Daging Sintesis Menggunakan Kultur Jaringan Sel Hewan

Pertumbuhan manusia di dunia mengikuti grafik eksponensial
yang terus bertambah dari tahun ke tahun. Populasi manusia
yang sulit dikontrol pertumbuhannya memunculkan isu
ketersediaan pangan dunia. Pertumbuhan ketersediaan pangan tidak
berimbang dengan pertambahan jumlah penduduk. Daging menjadi
salah satu jenis bahan pangan yang menjadi isu global. Oleh karena
itu, para ilmuwan mencari cara menghasilkan daging sebanyak
banyaknya dalam waktu singkat.
Perkembangan teknologi saat ini memungkinkan para ilmuwan
membuat daging melalui metode kultur jaringan sebagai alternatif
dalam produksi daging. Daging sintetis ini dihasilkan melalui teknik
in vitro.
Daging yang dikonsumsi umumnya terdiri atas serabut otot.
Sementara daging sintetis merupakan hasil pertumbuhan serabut
otot dalam kultur sel yang diambil dari sel punca atau sel induk
(stem cell). Daging sintetis ini dikenalkan untuk menjadi alternatif
pengganti daging konvensional.
Daging konvensional diperoleh dari seekor ternak yang
dipelihara selama periode waktu tertentu, kemudian dilakukan
proses pemotongan untuk memperoleh dagingnya. Proses produksi
daging konvensional saat ini dianggap memiliki pengaruh negatif
terhadap lingkungan, karena menghasilkan emisi gas rumah kaca
yang akan mencemari lingkungan.
Selanjutnya, pemotongan ternak dengan tujuan untuk
mengambil dagingnya diklaim sebagai suatu proses yang menyakiti
ternak dan disebut tidak memenuhi kriteria kesejahteraan hewan.
Di samping itu, timbulnya berbagai penyakit pada manusia seperti
jantung koroner, darah tinggi, kholesterol, dan stroke adalah deretan
penyakit yang disebabkan oleh mengonsumsi daging merah.
Daging sintetis ini diklaim sebagai daging yang sehat, ramah
lingkungan, tidak menyakiti hewan, serta menjadi solusi untuk
pemenuhan kebutuhan konsumsi dan produksi daging. Disebut
daging yang sehat, karena kandungan nutrien daging tersebut dapat
diatur sesuai kebutuhan serta dimodifikasi komponen lemaknya
sesuai dengan yang diinginkan. Produksi daging ini tidak melalui
proses pemotongan ternak karena sel induk dapat diambil dari
ternak hidup sehingga tidak menyakiti hewan. Produksi daging ini
juga dilakukan dalam bioreaktor yang hanya sedikit menghasilkan
gas metan sehingga disebut lebih ramah lingkungan.
Sebagai penemuan baru, nama yang diberikan masih beragam,
ada yang menyebutnya daging laboratorium, daging in vitro, daging
sintetis, daging bersih, daging Memphis (merujuk pada perusahaan
produksi) dan lain-lain. Selanjutnya, karena mengacu pada proses
asal daging yang diproduksi secara kultur jaringan, maka dapat
disebut daging sintetis.
Sumber Sel Induk
Untuk menumbuhkan daging dalam kultur diperlukan sel induk
(stem cell), yang dapat berasal dari embrio (embryonic cell) atau sel
dewasa (adult cell). Sel induk yang berasal dari embrio diperoleh dari
embrio yang belum menempel pada rahim, sedangkan sel induk
yang berasal dari sel dewasa diambil dari jaringan tubuh hewan
yang sudah lahir. Pada perkembangannya, sumber sel yang dapat
dipergunakan sebagai sel induk ini dapat diperoleh dari beberapa
sumber berikut ini:
Sel Satelit (Satellite Cells)
Satellite cells atau myosatellite cells adalah sel induk yang
diperoleh dari jaringan otot mamalia dewasa. Sel ini berfungsi untuk
regenerasi jaringan apabila terjadi kerusakan sel pada ternak hidup.
Percobaan pembiakan daging sintetis menggunakan sel satelit ini
sudah lama berhasil dilakukan di laboratorium. Proses pembiakannya
lebih mudah dilakukan dibandingkan dengan sel yang baru tumbuh.
Sel-sel satelit yang diperoleh dari teknik biopsi pada ternak hidup
atau saat sudah dipotong memiliki daya regeneratif yang terbatas
sehingga penggunaannya harus memperhatikan kesediaannya. Sel
ini tidak bisa disimpan lama sebagai bibit dalam kultur jaringan sel
hewan, sehingga setiap kultivasi harus mengambil bibit baru dari sel
hidup dengan cara biopsy. Dengan kata lain, ternak hidup masih
diperluka sebagai sumber sel induk setiap proses kultivasi.
Sel Induk Jaringan Adipose (Adipose Tissue-Derived Stem
Cells = ADSCs)
Adipose tissue-derived stem cells diperoleh dari stem cells
dewasa pada bagian tulang sumsum dan jaringan lemak. Jaringan
ini merupakan jaringan multipotent cell yang mampu melakukan
diferensiasi atau berkembang menjadi sel otot (myogenic), sel
pembentuk tulang (osteogenic), sel pembentuk tulang rawan
(chondrogenic), dan sel pembentuk lemak (adipogenic cell).
Penggunaan jaringan ini lebih mudah karena sumber sel dapat
diperoleh dari jaringan lemak subkutan (lemak di bawah kulit),
namun seperti halnya sel satelit, sel lemak sebagai bibit kultivasi juga
harus selalu dipanen dari ternak hidup.
Sel Induk Embrionik (Embryonic Stem Cells = ES Cells)
Embryonic stem cells dapat digunakan sebagai alternatif sumber
sel induk karena ES cells dapat memproduksi berbagai jenis sel seperti
pada janin dengan jumlah yang tidak terbatas, sehingga dapat
memproduksi jenis sel yang diinginkan. Kultur sel ini dapat disimpan
lama dan dibiakan sebagai bibit apabila diperlukan, sehingga tidak
memerlukan ternak sebagai sumber bibit setiap kultivasi. Namun,
untuk mengambil sel induk ini cukup sulit karena harus mengetahui
waktu yang tepat saat mengisolasi sel induk dari embrio. Pada saat
isolasi, kultur ES sulit dibedakan, apakah pada fase blastocysts atau
inner cell masses. Kesulitan selanjutnya adalah produksi ES cell line
pada in vitro menghasilkan embrio yang perkembangannya lebih
lambat dibandingkan embrio yang dihasilkan dari proses in vivo,
sehingga disarankan untuk menggunakan teknik in vivo dalam
menghasilkan embrio untuk sampel ES stem cells. Hal ini melibatkan
teknik pemanenan yang dapat mengganggu kesejahteraan hewan.
Induced Pluripotent Stem Cells (Sel Induk yang Diinduksi)
Induce pluripotent stem cells merupakan pluripotent stem cell
yang dibentuk dari manipulasi genetik atau epigenetik yang berasal
dari ekspresi sel-sel dewasa. Berbeda dengan sel-sel somatik atau
embryonic stem cells yang digunakan sebagai sampel dengan cara
biopsi. Menghasilkan pluripotent stem cells tidak memerlukan biopsi
maupun produksi dari fertilisasi telur. Sel ini dapat dihasilkan dari sel
apa saja dengan nukleus, seperti dari peripheral blood (sel darah)
atau skin cells (sel kulit).
Campuran Sel
Beberapa penelitian menggunakan sampel jaringan dengan
campuran sel sebagai pengganti dari sumber sel. Kelebihan
campuran sel ini di antaranya mengandung semua sel yang
membentuk otot dengan proporsi seimbang. Namun, juga memiliki
kekurangan di antaranya pertumbuhan substansial yang mungkin
tidak serupa pada beberapa jenis dari dimulainya kultur akibat dari
kekurangan sirkulasi darah. Sel dapat berasal dari kombinasi jenis
sel yang berbeda asalnya, bahkan dari beda spesies tetapi masih
memiliki karakteristik yang sama.
Cells for Other Components of Meat (Sel Induk dari
Komponen Daging Lainnya)
Sumber sel yang telah disebutkan memungkinkan untuk
menghasilkan daging, tetapi daging yang dihasilkan hanya terdiri
atas sel otot. Produksi daging agar memiliki struktur dan rasa seperti
daging asli masih memerlukan jenis sel lainnya, terutama jaringan
lemak (jaringan adipose). Co-culturing dari jenis sel yang berbeda
juga dapat memberikan manfaat untuk proses kultur tersebut, tidak
hanya untuk membuat rasa dan tekstur serupa dengan daging
asli tetapi juga karena co-culture dari jenis sel yang berbeda dapat
memberikan manfaat lainnya, berupa pasokan komponen lain untuk
pertumbuhan sel seperti vitamin, mineral, dan growth factor yang
lain.
Cara Membuat Daging Sintesis
Berikut ini adalah tahapan dalam membuat daging sintetis:
Pre-kultur (Ekstraksi)
Proses pre-kultur meliputi pemanenan stem cells dengan
cara biopsi. Stem cells yang sudah dipanen selanjutnya diisolasi
dan dipisahkan secara mekanik atau secara enzimatik, dilakukan
seleksi terhadap stem cells dan ditransfer ke media untuk
dikloning. Daging sintetis dapat disediakan melalui kultur stem
cell yang dibekukan setelah proses pre-kultur.
Kultivasi (Ploriferation)
Stem cells dari proses ekstraksi dipindahkan kedalam
media biakan yang umumnya disebut scaffolds. Scaffolds harus
bersifat biodegradable dan edible. Stem cells pada scaffolds
akan mengalami proses kultivasi sehingga tumbuh dan
melipatgandakan diri (poliferation).
Diferensiasi
Scaffolds yang sudah ditumbuhi stem cells kemudian
ditempatkan didalam bioreaktor dan dibiakkan pada media
pertumbuhan cepat. Proses diferensiasi meliputi pertumbuhan
dari stem cells menjadi myoblasts, myotubes dan myofibril.
Pembentukan Daging Sintetis
Sel yang telah mengalami diferensiasi kemudian ditempatkan
pada wadah vakum (cetakan). Sel akan tumbuh menjadi ribuan
muscle fibers (serabut otot) untuk memproduksi daging sintetis.
Pada gambar Skema dibawah dapat dilihat tahapan proses
pembuatan daging sintetis.
Keamanan Pangan Daging Sintetis
Menelusur faktor keamanan pangan produk daging sintetis
secara umum dapat dikatakan proses produksinya memenuhi
kriteria higienis sehingga ada yang menyebutnya daging bersih
(clean meat). Daging sintetis dapat dikonsumsi dengan aman dan
menyehatkan jika proses pembuatannya sesuai dengan standar
keamanan yang sudah ditetapkan.
Daging ini diproduksi dalam lingkungan yang steril, tidak
ditambahkan antibiotik, jumlah dan jenis lemak yang ditambahkan
juga dikontrol. Dengan demikian, daging sintetis akan bebas dari
mikroba, lebih sehat, dan ramah lingkungan.
Meski begitu, perlu kajian bersama antara ilmuwan dan ulama
untuk menelaah dan menghasilkan fatwa tentang kehalalan
daging sintetis, mengingat produk ini sudah mulai dilirik oleh para
pengusaha.
